Sugar Transport in Neurospora crassa*

چکیده
The transport of several sugars by cells of Neurospora crassa grown in shake culture has been investigated. 3-OMethyl-n-glucose is a substrate for the glucose transport system in glucose-grown cells but cannot be phosphorylated by the ATP-dependent hexokinase which readily catalyzes the phosphorylation of D-glucose. Since this nonmetabolizable glucose analogue cannot be accumulated against a concentration gradient in cells grown on glucose, it appears that net glucose uptake in glucose-grown cells involves a facilitated diffusion transport step followed by ATP-dependent phosphorylation and subsequent metabolism of the transported glucose. Growth of cells on D-fructose leads to the appearance of a system which catalyzes the accumulation of unaltered 3-O-methyl-D-glucose and L-sorbose against considerable concentration gradients.

Specificity of nucleoside transport in Neurospora crassa.
The specificity of nucleoside uptake in germinating conidia of Neurospora crassa was investigated by examining the kinetics of [2-14C]uridine and [8-14C]-adenosine uptake in the wild-type, ad-8, and ud-1 pyr-1 strains. The results obtained strongly indicate that nucleoside transport in N. crassa is mediated solely by a general transport system which accepts both purine and pyrimidine nucleoside...
متن کاملRegulation of sugar transport in Neurospora crassa.
Sugar uptake systems in Neurospora crassa are catabolically repressed by glucose. Synthesis of a low K(m) glucose uptake system (system II) in Neurospora is derepressed during starvation for an externally supplied source of carbon and energy. Fasting also results in the derepression of uptake systems for fructose, galactose, and lactose. In contrast to the repression observed when cells were gr...
متن کاملrco-3, a gene involved in glucose transport and conidiation in Neurospora crassa.
Macroconidiation in Neurospora crassa is influenced by a number of environmental cues, including the nutritional status of the growing organism. Conidia formation is normally observed when the fungus is exposed to air. However, carbon limitation can induce conidiation in mycclia submerged in an aerated liquid medium. A mutant was previously isolated that could conidiate in submerged culture wit...
متن کاملBiotransformation of Hydrocortisone by Neurospora crassa
The ability of Neurospora crassa FGSC 4335 in the biotransformation of hydrocortisone was investigated. The microorganism produced two major metabolites after incubation with the substrate for seven days. Each microbial product was purified chromatographically and identified on the basis of spectral data. The products were identified as 11?,17?,20?,21-tetrahydroxypregn-4-en-3-one (II) and 11?-h...
متن کاملSize restriction on utilization of peptides by amino acid auxotrophs of Neurospora crassa.
Growth of an amino acid auxotroph of Neurospora crassa on oligopeptides is shown to occur by extracellular hydrolysis, with subsequent utilization of monomer amino acid residues, and by transport of peptides. Peptides with a hydrodynamic volume greater than that of trileucine are not transported, and this lack of transport is shown to be due to restriction by the oligopeptide transport system r...
